Étude de la fonctionnalité des anticorps sériques et muqueux induits lors d’une vaccination ou d’une infection à Bordetella pertussis, l’agent de la coqueluche

Abstract : Most studies in the pathogenesis of Bordetella show persistent colonization of the bacterium in the lower respiratory tract, but recent researchs have shown that it can colonize the upper part of the respiratory tract, including the nasal cavity. These bacteria may induce local mucosal immunity, involving the potential role of secretory IgA in local control of infection. First, we plan to develop and use a high throughput growth inhibition test. This test, which we called BGIA (Bordetella growth inhibition assay), will evaluate the bactericidal and bacteriostatic properties of various biological fluids collected at different times of bacterial colonization after infection or vaccination with B. pertussis in mice, baboons and humans (serum, nasal wash and bronchoalveolar). In a second step, the inhibitory molecules (antibodies, comlement, peptides) and inhibitory mechanisms observed will be studied in greater depth. With this thesis project, we hope to advance on one hand in understanding the protective mechanisms of the humoral response and on another hand the relationship between local immunity and systemic immmunity.
